What the Substitutions Do

Native GHRH is cleaved within about two minutes in plasma by dipeptidyl peptidase IV (DPP-IV), which removes the N-terminal dipeptide. The D-alanine at position 2 blocks that cleavage; the other three substitutions were introduced for structural stability of the peptide. The animal-model pharmacokinetic literature summarised in the comparison guide reports a plasma half-life in the order of 30 minutes for the no-DAC sequence, against days for the DAC-linked variant, which travels bound to albumin.
